Dental methacrylates may exert genotoxic effects via the oxidative induction of DNA double strand breaks and the inhibition of their repair

Abstract

Methacrylate monomers used in dentistry have been shown to induce DNA double strand breaks (DSBs), one of the most serious DNA damage. In the present work we show that a model dental adhesive consisting of 45% 2-hydroxyethyl methacrylate (HEMA) and 55% bisphenol A-diglycidyl dimethacrylate (Bis-GMA) at concentrations up to 0.25 mM Bis-GMA induced oxidative DNA in cultured primary human gingival fibroblasts (HGFs) as evaluated by the comet assay and probed with human 8-hydroxyguanine DNA-glycosylase 1. HEMA/Bis-GMA induced DSBs in HGFs as assessed by the neutral comet assay and phosphorylation of the H2AX histone and sodium ascorbate or melatonin (5-methoxy-N-acetyltryptamine) both at 50 μM reduced the DSBs, they also inhibited apoptosis induced by HEMA/Bis-GMA. The adhesive slowed the kinetics of the repair of DNA damage induced by hydrogen peroxide in HGFs, while sodium ascorbate or melatonin improved the efficacy of H2O2-induced damage in the presence of the methacrylates. The adhesive induced a rise in the G2/M cell population, accompanied by a reduction in the S cell population and an increase in G0/G1 cell population. Sodium ascorbate or melatonin elevated the S population and reduced the G2/M population. In conclusion, HEMA/Bis-GMA induce DSBs through, at least in part, oxidative mechanisms, and these compounds may interfere with DSBs repair. Vitamin C or melatonin may reduce the detrimental effects induced by methacrylates applied in dentistry.

Zusammenfassung

Methacrylatmonomere, die in der Zahnmedizin verwendet werden, induzieren nachweislich DNA-Doppelstrangbrüche (DSBs), eine der schwerwiegendsten DNA-Schäden. In der vorliegenden Arbeit zeigen wir, dass ein Modell-Dentaladhäsiv, das zu 45 % aus 2-Hydroxyethylmethacrylat (HEMA) und zu 55 % aus Bisphenol A-Diglycidyldimethacrylat (Bis-GMA) besteht, bei Konzentrationen von bis zu 0,25 mM Bis-GMA oxidative DNA in kultivierten primären humanen Zahnfleischfibroblasten (HGFs) induziert, wie mit dem Comet-Assay bewertet und mit humaner 8-Hydroxyguanin-DNA-Glykosylase 1 untersucht wurde. HEMA/Bis-GMA induzierte DSBs in HGFs, wie durch den neutralen Comet-Assay und die Phosphorylierung des H2AX-Histons bewertet wurde, und Natriumascorbat oder Melatonin (5-Methoxy-N-Acetyltryptamin), beide in einer Dosierung von 50 μM, reduzierten die DSBs und hemmten auch die durch HEMA/Bis-GMA induzierte Apoptose. Der Klebstoff verlangsamte die Kinetik der Reparatur von durch Wasserstoffperoxid induzierten DNA-Schäden in HGFs, während Natriumascorbat oder Melatonin die Wirksamkeit von H2O2-induzierten Schäden in Gegenwart der Methacrylate verbesserten. Der Klebstoff führte zu einem Anstieg der G2/M-Zellpopulation, begleitet von einer Verringerung der S-Zellpopulation und einem Anstieg der G0/G1-Zellpopulation. Natriumascorbat oder Melatonin erhöhten die S-Population und reduzierten die G2/M-Population. Zusammenfassend lässt sich sagen, dass HEMA/Bis-GMA DSBs zumindest teilweise durch oxidative Mechanismen induzieren und dass diese Verbindungen die Reparatur von DSBs beeinträchtigen können. Vitamin C oder Melatonin können die schädlichen Auswirkungen von Methacrylaten, die in der Zahnmedizin verwendet werden, verringern.
